- Collector qualifications: Urine sample collectors must complete qualification training and demonstrate proficiency by completing five consecutive error-free mock collections under 49 CFR Part 40 before conducting DOT testing
- Collection site standards: Sites must include a private room with a door and one toilet; water sources must be secured before collection begins to prevent tampering
- Chain of custody: Every individual who comes into contact with a specimen must be trained on proper DOT protocols and must sign for the sample at each transfer point
- SAMHSA-certified laboratories: Laboratories used for DOT testing must be certified by the Substance Abuse and Mental Health Services Administration
- Medical Review Officer (MRO) review: An MRO determines the final verified result, including whether a specimen has been tampered with or is diluted, and reviews the employee’s prescription history before issuing a result
- Error correction: If collection or documentation errors occur, a test can be cancelled and the employer may face noncompliance exposure; error correction training is required within 30 days of being notified of a mistake
